Were you able to confirm your ODB2 dongle is appearing as Com1 in device manager? Did you set the latency to 1 in settings? When you start inpa connected to the car are the 2 indicator circles for cable found and ignition on checked?

As for ISTA, that should work with no ODB2 cable attached, obvs not to diagnose but to display the TIS procedures and diagrams. Not sure if can't find the sqlite databases or finds them and can't open them for some reason. I'm wondering if when you copied the program and started it from the tools folder it wrote some registry entries for the wrong path. Maybe try downloading CCleaner and running the registry cleaner.
In the Rheingold folder there is a folder named Logs, in that should be a log file with the name being the date, your pc name and ista, try to attach that log file or copy it into a post.

I just installed that version on my desktop with no dongle or drivers or anything and it works when I enter a VIN. Interestingly this version will only accept last 7 of VIN, the version I have on my diagnostic laptop will only accept the full VIN.

I believe the two lights are showing that it sees the cable and ignition on.

Ill try and run the registry cleaner on my laptop when i get home. Ill let you know results after doing these. thank you
