Does DISA flaps work on 325i with 3 stage manifold installed without any tuning?

No. The wiring is pre-installed tho. If you want the DISAs to work you need a tune. The free Bimmerlabs 325ito330i tune is a great choice, especially on stock exhaust pipes and manifold.

After installing the 330i tune can I still revert to stock tune later? I also noticed there is an RSA delete file from bimmerlab website. Not sure if this will cause any error code that pops up every time...

Yes you can always revert it to stock. I'm running that tune and the only error is a undefined DME error "No Coding". It doesn't do any harm tho, no lights or module/sensor errors or anything. hassmaschine the creator says it's just a phantom error some kind of artifact from the tune but not doing or meaning anything.
